Endophytic Microbiome Variation Among Single Plant Seeds

چکیده
Like other plant compartments, the seed harbors a microbiome. The members of microbiome are first to colonize germinating seedling, and they may initiate trajectory assembly for next generation. Therefore, important dynamics vertical transmission potentially beneficial symbionts. However, it remains challenging assess at individual level (and, therefore, future plants) due low endophytic microbial biomass, exudates that can select particular members, high plastid contamination resulting reads. Here, we report protocol extracting DNA from an (common bean, Phaseolus vulgaris) with minimal disruption host tissue, which expect be generalizable medium- large-seed species. We applied this determine 16S ribosomal RNA (rRNA) V4 rRNA internal transcribed spacer 2 amplicon composition examine variability harvested replicate common bean plants grown under standard, controlled conditions maintain health. Using extractions seeds, compared seed-to-seed, pod-to-pod, plant-to-plant microbiomes, found highest level. Search for endophytic diazotrophs in barley seeds
Eight endophytic isolates assigned to Pseudomonas, Azospirillum, and Bacillus genera according to pheno-genotypic features were retrieved from barley seeds under selective pressure for nitrogen-fixers. Genetic relationships among related isolates were investigated through RAPD. Six isolates displayed nitrogen-fixing ability, while all could biosynthesize indolacetic acid in vitro and showed no ...
متن کاملWhat Is There in Seeds? Vertically Transmitted Endophytic Resources for Sustainable Improvement in Plant Growth
Phytobeneficial microbes, particularly endophytes, such as fungi and bacteria, are concomitant partners of plants throughout its developmental stages, including seed germination, root and stem growth, and fruiting. Endophytic microbes have been identified in plants that grow in a wide array of habitats; however, seed-borne endophytic microbes have not been fully explored yet. Seed-borne endophy...
متن کاملIdentification of predominant epiphytic and endophytic bacterial isolates in rice seeds effective for enhancement of seed germination and plant growth
ABSTRACT-During rice growing season, the symptomless rice seeds from different paddy fields in Guilan province, Iran, were collected. After isolation of epiphytic and endophytic bacteria, 39 isolates including 19 epiphytes and 20 endophytes were selected based on the predominant characteristics. Five Operational Taxonomic Units (OTUs) were obtained based on PCR-RFLP of 16S r-DN...
متن کاملThe Plant Microbiome
How do we define the ‘plant microbiome’ and what is its significance to the plant genome? Before addressing what the microbiome is in relation to plants, it is important to first understand the concept of the microbiome; what this means in relation to the host becomes an extension of this working concept. Conceptualizing the microbiome requires a fusion of microbial ecology and bioinformatics, ...
متن کاملEndophytic fungi from flowers, capsules and seeds of Eucalyptus globulus.
The main goal of this work was to detect whether Cytospora chrysosperma and Fusicoccum eucalypti are present as endophytes of symptomless hypocotyls, cotyledons, flowers, capsules, peduncles of flowers in order to interpret an earlier finding of their presence in seeds, seedling stems and twigs of E. globulus. Segments from these organs as well as from bark and the xylem from flower peduncles w...
